Beta-caryophyllene (BCP) occupies a unique position in plant chemistry: it is the only terpene known to directly bind to cannabinoid receptors in the human endocannabinoid system. Specifically, it acts as a selective agonist at the CB2 receptor, which is expressed primarily in immune cells, the spleen, the gastrointestinal tract, and the peripheral nervous system. This dual identity — simultaneously a terpene and a functional cannabinoid — earned it the designation as a "dietary cannabinoid" in a landmark 2008 paper published in the Proceedings of the National Academy of Sciences.

The CB2 receptor system is fundamentally different from CB1, the receptor through which THC produces its psychoactive effects. CB2 activation does not produce any "high" or altered consciousness. Instead, it modulates immune function, reduces inflammatory signaling, and influences pain perception at the peripheral level. When Caryophyllene binds to CB2, it triggers a cascade of anti-inflammatory responses that include reducing production of pro-inflammatory cytokines (TNF-alpha, IL-1beta, IL-6), inhibiting inflammatory cell migration, and downregulating inflammatory gene expression through the NF-kB pathway.

This CB2 binding capability sets Caryophyllene apart from every other terpene in the cannabis plant. This means it contributes to the entourage effect not just as a supporting player, but as an active participant in cannabinoid receptor signaling.

The molecular structure of Caryophyllene — a bicyclic sesquiterpene with a rare cyclobutane ring — is what enables this receptor binding. The cyclobutane ring is unusual in natural compounds and gives Caryophyllene a three-dimensional shape that fits into the CB2 receptor binding pocket with moderate affinity. Its binding is selective enough that it does not meaningfully interact with CB1, explaining why Caryophyllene supplementation (even in large doses from dietary sources like black pepper) does not produce psychoactive effects.
